The invention relates to a preparation method of targeted long-circulating nano drug carriers for photothermal synergistic chemotherapy. The preparation method comprises the following steps: preparing mesoporous silica; coating mesoporous silica with polydopamine; loading anti-cancer drugs; modifying erythrocyte membranes with biotin; wrapping the drug-loaded mesoporous silica with biotin-modified erythrocyte membranes. The present invention realizes local photothermal/chemotherapy synergistic treatment of cancer tissues through polydopamine (PDA) coating to improve treatment efficiency. Wrapping with erythrocyte membranes enables the synthesized nanoparticles to effectively evade biological barriers, circulate in the whole body, and achieve high accumulation in tumors. Active targeting is realized by biotin modification to further enhance aggregation at tumor sites, which greatly improves the biocompatibility of drug carriers and their circulation time in human bodies.
